چکیده

To improve the characterization of electrophoretic lipoprotein subfractions, we developed two new techniques for analyzing lipoproteins after electrophoresis on thin agarose layers. Overlay with antisera exactly localizes specific apoproteins without any distortion caused by antigen diffusion; electron microscopyof eluted fractions determines the varying particle-size distribution. Applied together, these methods can detect individual differences between hyperlipemic samples that are not immediately apparent in the electrophoretic pattern, and should provide valuable new insight into the classification of hyperlipoproteinemias.

Investigation of the abnormal low-density lipoproteins occurring in patients with obstructive jaundice.

The abnormal lipoproteins of the density range 1.019-1.063g/cm3 occurring in the plasma of patients with obstructive jaundice were studied. Subfractionation of this density class by combined sodium phosphotungstate precipitation, ultracentrifugation, and column chromatography on hydroxyapatite and agarose gel yielded essentially three fractions: (1) lipoprotein-X, (2) A triglyceride-rich lipopr...

Isolation and characterization of lipoprotein B from high-density human serum lipoproteins.

1. Lipoprotein B from female Lp(a)-lipoprotein-negative serum was isolated from the fraction of density 1.073-1.125 by using immunoadsorbent; 2.5mg of freeze-dried material was obtained from 100ml of serum from a fasting patient. 2. The hydrated density of this lipoprotein was found to be 1.084g/cm(3). A flotation rate F(1.200) of 9.4 and lipid/protein ratio 1.40 were found, similar to that of ...

Changes in rabbit lipoprotein properties by dietary cholesterol, and saturated and polyunsaturated fats.

Plasma lipids and chemical, electrophoretic and electron microscopic properties of VLDL, LDL and HDL are examined in rabbits fed a control diet (group I) or diets containing 1% cholesterol (group II), 1% cholesterol + 5% coconut oil (group III) or 1% cholesterol + 5% corn oil (group IV). The diets II, III and IV resulted in hypercholesterolemia, hypertriglyceridemia and hyperphospholipidemia. T...

Isolation and characterization of electrophoretic variants of human prostatic acid phosphatase.

Prostatic acid phosphatase (EC 3.1.3.2) purified from benign hypertrophic prostate tissue was fractionated by preparative slab isoelectric focusing over a pH gradient of 3.16 to 7.16. Twenty-two of 29 fractions contained enzyme activity. We further examined each active fraction by determining the Michaelis-Menten constant and specific activity. The protein concentration used in the latter deter...

Lecithin--cholesterol acyltransferase and the lipoprotein abnormalities of obstructive jaundice.

1. We have studied the plasma lipoprotein abnormalities in obstructive jaundice to test the hypothesis that the abnormalities would correlate with plasma lecithin--cholesterol acyltransferase activity. 2. Very-low-density lipoproteins (VLDL) were normal in composition and had a normal pre-beta electrophoretic mobility when lecithin--cholesterol acyltransferase activity was high. When it was low...
